1) Can I use two softphones(Linophone and Ekiga) on my machine to
simulate communication over SER?

No. It's not at all possible, you must configure each aplication (ser,linphone,ekiga) to use diferent ports.

3) Do I need to setup mediaproxy in this case since both phones are on
my machine?

No. simplest config file will make the job.
